Your section on sampling should mention the sealed vial. A laboratory that receives a factory-sealed container and breaks the seal itself can say something about what was in the container. One that receives a decanted aliquot in a plain tube can say something about the tube.

Your description of collapse is right and the consequence is worth spelling out. A collapsed cake has a lower surface area and reconstitutes more slowly, and it also indicates the product went above its collapse temperature during primary drying. It is a visible record of a process deviation, sitting in the vial where anybody can see it.

We can say where it comes from, which is the in-use period established for licensed pen presentations of specific formulations in specific containers. Whether it transfers to a different peptide reconstituted in a different diluent in a different vial is not something the stability literature permits anyone to assert. I work in a contract laboratory and I would note that some data systems export a representative overlay rather than the individual injection, so identical-looking traces can occur legitimately across a batch series. It is still worth asking about; it is not the smoking gun your article implies.

Water content by Karl Fischer is inexpensive, fast and almost never reported. For a lyophilised powder it bears directly on stability, and it is the determination I would add first if I could add only one. Material described as being for research use only is not approved for human use in any case, so the argument for measuring it is a scientific one rather than a safety one.

Your worked example separates two species one dalton apart and treats that as the hard case. The hard case in practice is fourteen milli-daltons, which is what distinguishes a genuine methyl group from a substitution nobody wanted. That gap needs an order of magnitude more resolving power than most service instruments deliver.

A caution on the word high-resolution as it appears in marketing. It is not a threshold; it is a continuum, and instruments separated by a factor of twenty in resolving power are sold under the same adjective. Ask for the figure and the mass at which it was obtained, and the adjective becomes unnecessary.

Ask for the acquisition method file, not the report. The report is a rendering of a decision; the method file is the decision. Two laboratories will send you the same one-page summary from acquisitions that have almost nothing in common.

Your article says a matching mass does not confirm a sequence, which is correct, and then rather implies that vendors are trading on the ambiguity. I run analytical services and I would put it differently: we report what we measured, in the words our clients ask for. If the Journal wants the word confirmed retired, write to the buyers, not to us.

That is a fair reallocation of the criticism and we accept it. The word is chosen by whoever commissions the report, and laboratories are answering the question they were paid to answer. Our complaint is with the practice, not with the analysts, and the article should have located it more precisely.

The instrument-class section is the most useful thing you have printed on this subject. A unit-resolution quadrupole cannot distinguish a deamidation from the parent, and a great many identity confirmations in this trade are performed on exactly that class of instrument. The report says the mass matched, and it did, at the resolution available.

The number worth asking for is resolving power at the mass you care about, not the headline figure from the brochure. Instruments are specified at a mass convenient to the manufacturer, and a peptide at four thousand daltons is not that mass. Ask where the specification was measured.

The former, which the table note now says explicitly. The count for what appears on a customer-facing certificate is lower in at least four cases, and we should have separated the two columns rather than merging them.

Two ionisation techniques on the same sample is the cheapest confidence a laboratory can buy. Electrospray and matrix-assisted desorption fail in different directions, and a species that survives both is far more likely to be real than one that appears only in the technique the laboratory happens to own.
